The manga and the anime series differ in Wolfwood's history, and that's not the only thing they differ in. In my personal opinion, the manga totally rips the anime series apart like day old donuts that have been out in the sun too long. The manga totally rocks and compaired to that, the series really falls short. It's a shame...

ANIMATED SERIES

In the anime series, Wolfwood's parents were killed by his guardian and was sent to the church/orphanage outside of the city of December. When he reached the age of seven, he shot the murderer of his family who was his guardian. Nick then meets up with Chapel the Evergreen whom teaches him most of everything Wolfwood knows: how to shoot, how to build the Punisher, and even his outlook on life including his religious standpoints. It's said that Wolfwood became a preacher after his mentor, Chapel who was a Gung-ho Gun. Wolfwood looked up to Evergreen so much that he walked in his footsteps and became a Gung-ho Gun himself, earning the feared title of "The man who rings the black funeral bell."
Because he had such a crappy childhood, he swore that he would always protect and give hope to those children that had lost their way, much like how Chapel gave him direction. He evetually claims the church outside of December as his own, or maybe it was given to him by Evergreen. He can't make enough money for the children he protects by being a traveling preacher, so he decided to make some...money by doing work as a Gung-ho Gun.
Wolfwood was secretly assigned to protect and guide Vash to Knives and not even Legato knew about it. I figure that Knives had a lot of faith in the preacher, maybe even more than he did in Legato because he gave Nick this important mission and didn't tell anyone about it. All of the Gung-hos suspect that Wolfwood is a Gung-ho, and this scares them a little bit because they have heard stories of his slaughtering.
In the series, Wolfwood seemingly accidently runs into Vash in the middle of the desert because his "Shiny Angelina II" broke down and he had to travel by foot. Vash spots him in the far off distance from the glint of the sunlight reflecting off of the silver buckles of the Punisher. They slowly become friends that convinces Wolfwood to change his plans from killing Vash and Knives to just killing Knives. Though even near the end, Wolfwood struggles to not kill Vash.
After many hardships and saving Vash's ass and the insurances girl's many times, he is forced into his final confrontation with his mentor, Chapel the Evergreen. Wolfwood beats him but doens't kill him. This may be because Vash's wussy pacifism rubbed off on Wolfwood, but I think it is because Chapel was Wolfwood's mentor and he wouldn't have killed him anyway. As Wolfwood turns to walk away from Evergreen, Evergreen is forced to shoot him in the back by Legato's influence. Dying and bleeding horribly, Wolfwood drags himself into a local church where he confesses his sins for the very first time, despite his profession. Soon afterwards he passes away. (Personally I don't think he is really dead, but I think it was a very good scene. I cried, and I am man enough to admit it.) Vash's life is saved for a final time by the good minister as the Punisher was used by Vash to beat Knives in the final confrontation.

MANGA

In the manga, Wolfwood IS Chapel. I've heard from different sources that his name is Chapel the Peacemaker and others say he is Chapel the Evergreen. Either way, he IS Chapel. The Chapel in the animated series was a fake character that was made up for no other reason than to be annoying, and to die like a chump.
In the Manga, Wolfwood and Midvalley team up and conspire to kill both Vash, Knives and Legato. Just like in the series, Wolfwood grows fond of Vash and changes his plans.
Wolfwood has no strong feelings for Millie, not even luke-warm feelings for the insurance girl in the manga. Which I find more appealing anyway, well, maybe not. It just depends on my mood.
Other than the fact that Wolfwood kicks much more ass in the manga, and that he IS STILL ALIVE, PRIASE THE LORD!, there isn't much else different between the Manga and the Series conserning Wolfwood.
